Marvel's Spider-Man: After being bitten by a genetically modified spider, Peter Parker gains spider-like powers and can accomplish superhuman activities. Initially using his powers for petty crimes, Peter inadvertently causes the death of his uncle and forever paralyzes bully Flash Thompson. Intrigued and traumatized, Peter takes up the secret life of a crimefighter in an alter-ego entitled Spider-Man.
In a ferocious battle with the Russian war criminal Aleksei Sytsevich, Peter 's identity is accidently revealed to high school nerd Miles Morales, who has a record of mental depression and distress. Morales, wanting to feel special like Peter, attempts to develop a Spider-Man serum which goes horribly wrong. Testing on himself, Morales becomes a man hybrid of a ferocious lizard. Peter must put an end to the rogue Lizard who is causing mishap everywhere. By the end of the movie, the Lizard kills Captain George Stacy and fatally injures Gwen Stacy. Desperate, Peter tranfuses his blood to Gwen, and that gifts Gwen spider-like powers as well. Morales is cured as the Lizard and escapes, staying in low profile. Spider-Man's vigilancy is reduced as well. Gwen decides to honor his father's noble intentions and become a crimefighter as well, Spider-Woman. This way, Gwen can have her own spin-off series.

After being abandoned by his parents at a young age for mysterious purposes, Peter Parker, is left to his aunt May and uncle Ben Parker, and copes and struggles to blend in with the society, although being the main source of playfulness for the bullies, especially Flash Thompson. Peter also develops a strong friendship with Harry Osborn, the distressed son of the infamous Norman Osborn, multimillionare CEO of Oscorp Industries. He also develops an unoblivious crush on the popular and charismatic Gwen Stacy. Peter also grows the highest academic marks on Midtown High School
In his 17th birthday, Peter is publicly embarrased by Flash Thompson and his gang of bullies. Frustrated, Peter swears vengeance and puts all of his frustration and anger towards his aunt and uncle.
In order to calm and relieve Peter, Harry invites him to Horizon Labs, an Oscorp test facility to meet his childhood idol, Dr. Otto Ocktavius, who Peter has always idolized and looked up as a father figure. Otto Octavius, who dreams of perfecting sustained fusion power to create a portal, and wears a harness of powerful robotic tentacle arms with artificial intelligence while conducting his research. Before the beginning of the experimentation of nuclear fusion, Peter sees that Norman Osborn acting suspicious and decides to follow him, before accidently entering a restricted research zone. Otto begins his experiment. Within the research zone, Peter inadvertently enters a quarantine zone where he gets mutated by a biocable radioactive arachnid who passed on it' symptoms to Peter before death. Peter, manifesting spider like abilities and in pain, causes a radioactive explosion and escapes the building in confusion and terror.
Meanwhile, because of the explosion Peter caused, Otto's nuclear fusion goes absolutely wrong and the result causes the demise of Otto's wife, Norman Osborn sucuumbs to fatal injuries, many people get injured and Otto gets sucked into a rogue portal that is created by the accelerated nuclear fusion. Getting sucked into another dimension, Otto and his mechanical tentacles merge with him and some otherwordly elements combine with him to completely alter his physique, but somehow Otto gets out of the other dimension into the destroyed Horizon Labs place once again by a reverse portal.
Midst, Peter, afraid of his powers, lashes out in Hell's Kitchen, where he angers many goons and starts a fight with them, Peter defeats them and provokes Luke Cage, aka Power Man, the guardian of Hell's Kitchen, to have a brief duel with him. Luke Cage calms Peter down and mentors and resorts him to a fair point, and tells him to do whatever he wishes to do. Peter returns home and after experiencing his most twisted day ever, Peter realizes he can use his powers for his own personal gains and obsessions. Four weeks has past, Peter has now created a fugitive alter-ego, which he uses for crimes and beating up bullies. Peter increasingly becomes popular and more arrogant because of his strength in high school. He disputes with Harry because of his newfound fame and arrogance. He also starts dating Gwen Stacy, who later also breaks up with him because of his utter disrespect for anyone.
When he realizes that his relationship with everyone is dwindling apart, Peter grows the desire to murder Flash, the thing he wanted to do since he first got bullied. Peter donning his mask in his alter-ego spots Flash and takes him to an alley where he attempts to kill him, before being interrupted by uncle Ben who was following Peter. Peter panics and realizes of all of the wrong things he has done. Panicking and having a nervous breakdown, Peter causes a radioactive explosion, paralyizing Flash, and killing Uncle Ben. Terrorized, Peter escapes.
At a hospital, doctors prepare to surgically remove Octavius' arms and harness, but the arms, having developed sentience from the inhibitor chip's destruction, spring to life and attack the medical crew, killing most of them. Upon regaining consciousness and seeing the carnage, Octavius escapes and hides at a harbor. The arms convince him to retry the experiment. To fund it, Octavius – now called Doctor Octopus by the Daily Bugle – robs a bank.
Peter realizes that the fugitive and criminal, dubbed the Doctor Octopus, is actually Otto Ocktavius and all of the new disastrous occurences have been caused by him. In order to be clear of sins and guilt and deciding to end what he started, Peter creates an alter-ego, Spider-Man, in order to stop the mayhem caused by Doctor Octopus and also erase suspicion of being the vigilante that killed Uncle Ben. Peter is recovering all his lost aspects of his life, including financial problems, friendship problems, disputes, and also getting a job at the Daily Bugle as a photographer.
After Peter misses dozens of dates with Gwen, he is given a last chance with Gwen for a date, but he instead helps a woman from being harrased by a gang. Gwen breaks up with him for being late again, and moves on, and starts dating Harry. Peter suffers an emotional breakdown causing him to believe he's lost his powers. He abandons his Spider-Man identity and returns to his normal life while trying to reconcile with Gwen.
A garbageman brings Spider-Man's costume to J. Jonah Jameson, who takes credit for Spider-Man's disappearance. Peter tells Aunt May that his Uncle Ben's death some time ago was his fault. May forgives him, but when his 9-year-old neighbor learns of Spider-Man's disappearance (who is not actually a superhero but a symbol of hope) and the subsequent rising crime rate in New York City, Peter becomes concerned.
Octavius needs tritium to fuel his reactor and goes to the wounded Norman Osborn to demand it. Norman initially refuses because the experiment threatens to level the city, but he eventually agrees in exchange for Spider-Man who exposed the darkest secrets of Oscorp to the public and tells him that Peter, Harry's best friend, who is supposedly good friends with Spider-Man, is the key to finding him. Octavius finds Peter, tells him to find Spider-Man, and abducts Mary Jane. Peter realizes his powers are restored due to the trauma of seeing Mary Jane kidnapped, and dons his costume again after stealing it from the Bugle.
As Spider-Man battles Octavius, they fall onto a rapid transit R train. Octavius disables the controls and jumps off, but Spider-Man stops the train before the tracks end. When he faints from exhaustion, the passengers carry him into one of the cars. He comes to and realizes his mask is off, but the passengers are so grateful they vow not to reveal what he looks like. Octavius returns, demanding Spider-Man, and subdues the passengers. After knocking out Spider-Man, Octavius delivers him to Norman.
After giving Octavius the tritium, Norman prepares to kill Spider-Man in anger, but Peter (still masked) becomes involved in a duel fight with Norman. Peter discovers that Doctor Octopus intends to renew his experiment, and that is why, in need to stop Doctor Octopus, Peter reluctantly and without any option, causes a radioactive explosion that completely wounds Norman Osborn, and leaves him in a critical condition, and learns of Doctor Octopus' location.
Spider-Man arrives at the doctor's waterfront laboratory and tries to rescue Gwen discreetly. One of Octavius' tentacles senses him, and they fight. Spider-Man ultimately subdues Octavius, reveals his identity, and convinces Octavius to let go of his dream for the greater good. Octavius finally commands the tentacles to obey him and drowns the fusion reactor, along with himself, in the Hudson River. Gwen discovers Spider-Man's true identity, as well as why they cannot be together.
After the battle with Doctor Octopus, Peter realizes that he can probably be forgived of his sins by devoting his life to complete justice. Also, as New York is in need of a guardian, Peter decides to permanently adopt the mantle of Spider-Man, now as a guardian, but also as a symbol.
Gwen also breaks up with Harry, and reconciles with Peter, and tells him that her father is a NYPD captain, and like him, she wants to live all the adventures, and she wants to live all the adventures with Peter. With everything back to normal, Peter as the Spider-Man swings through the city before he foils a bank robbery by the Shocker, and the movie ends and the scene cuts to black before Peter and the Shocker engage in combat (like the ending of TASM2).
In a mid credits scene, Nick Fury and agent Sharon Carter, secretly enter the area in Horizon Labs where the disastrous events occured, and the portal was opened. They in hazmat suits augment the extraterrestrial elements that came out of the otherdimensional portal from which Otto popped out of, foreshadowing the creation of Man-Thing (in this universe, SHIELD was responsible for the Man-Thing).
In a post credits scene, Norman Osborn, in an isolated hospital room, suddenly opens his eyes which turns green, and his nerves become visible, indicating his later transformation to the Green Goblin and his involvement with later Spider-Man films.
