Agamotto is the first human to ever master the Mystic Arts. Beginning to travel the world attempting to recruit worthy students, she quickly learned that most of humanity could not be trusted with the truth. This was especially true of the Time Stone, which she had been able to summon through a series of chronological spells.
Unwilling to tamper with the Stone's power, Agamotto began drawing power from the Dark Dimension to live for thousands of years. Eventually, she was able to set up shop in eastern India, in a rumored place called Kamar-Taj. Over the years, she was able to put together a small society of trustworthy students with adept talents for the Mystic Arts.
Her most trusted lieutenants were Arthur Wong and Karl Mordo. Eventually, she began to suspect Mordo of colluding with Dormammu, making her hesitant to trust anyone. That is why, when Stephen Strange arrived, she initially refused to teach him.
However, after he was able to gain entrance into Kamar-Taj, she tasked Wong and Mordo with taking over his training so that she could observe them. Over time, she began to trust Strange, seeing the potential he held. But this only sped up Mordo's inevitable betrayal. His alliance with Dormammu resulted in a tear in reality, which the Masters of the Mystic Arts were forced to rally against. In the process, Mordo revealed the truth about the Ancient One, and how she channeled energy from the Dark Dimension to survive. Wong and Strange were shocked and heartbroken, but still tried to save her after Mordo severed her connection. But it was all in vain, as Dormammu was then able to finally kill the Ancient One, turning over the title of Sorcerer Supreme and possession of the Time Stone to Doctor Strange.
